For PS3, I would say Borderlands, as I have yet to get my hands on Demon's Souls due to it not being released in the UK yet, and the prices on eBay are just stupid.
For the iDevice, I love Rolando 2, and I think it is unappreciated, especially compared to Soosiz, which I seriously don't understand the love for.
As for the PC, Diablo, WoW, Starcraft.. anything made by Blizzard.
Mass Effect is good for 360.
Sonic Adventure 2 is my favourite for the Dreamcast.
Shadow of the Colossus and Baldur's Gate 2 are excellent on the PS2, but SotC takes the Cake.
As far as Classics go, Pac-Man is the best.
Super Smash Bros. Brawl is my personal favourite for the Wii.
The Pokemon Franchise is the best for GB/GBC/GBA/DS.
Ocarina of Time is the best for N64.
Ristar is my favourite from the SEGA Genesis.

However, I Wanna Be The Guy is the best game I have ever played.

Tough call... I have different favourites for different reasons.

Jet Set Willy, because it was the first game I ever played and therefore gives me such strong vibes of nostalgia that it's not physically possible to get bored of it.

Super Mario World, for taking the best of every previous Mario game, combining them together and then multiplying it by a thousand.

Ninja Gaiden Black/2, for having the most badass combat mechanics in the history of gaming. There aren't many games that let you pwn the enemies with such a unique combination of brutality and elegance.

Goldeneye, for being the first console FPS worth talking about, way before Halo sneaked in and tried to pretend it'd never been done before.

FFVII, although it may not technically be the best FF game ever, it holds the fondest memories because I always played it with my best friend, and sometimes the atmosphere you have playing a game is more important than how good the game itself is.

There are a bunch more, but... y'know... it'll go on for too long.

Morrowind, with all its flaws, I'm replaying it right now, great way to spend the holidays.

I got mine for £33 on eBay, cheaper than it'd be if it were out over here. Hint: get the Hong Kong version.

It's because you have chosen what your new favourite game will be before you've played it.

These are wise words, and with that, my favorite game is probably Final Fantasy VI. I also have very fond memories of this game that make up for any problems with the gameplay or story, which honestly, are mostly related to how old it is.

But it's a tough call. Final Fantasy VI holds the largest spot in my heart, but there have been many other games that are just as good: TIE Fighter, RE4, Shining Force (Another nostalgic one), Metal Gear Solid 3, and so on.

    The list goes on.

BEST GAME EVER: Metal Gear Solid. If you see how many other awesome games I have up here, it really shows how truly great this game is. Crisis Core comes in second, just needed to mention that.

NES: Earthbound Zero and Metal Gear

SNES: Super Mario RPG and Earthbound

Genesis: Mortal Kombat (it's kinda a nostalgia thing... used to play it alot when i was younger)

PSX (other than MGS): Final Fantasy VII

    N64: Paper Mario and Banjo-Kazooie

Gamecube: Resident Evil 4 or Custom Robo

PS2: Metal Gear Solid 2: Sons of Liberty and Final Fantasy X

Xbox: Doom 3 (although I played it on PC) or Tony Hawk's Underground (only one I've played on the Xbox, but my second favorite in the series)

Wii: Call of Duty: Modern Warfare: Reflex or Resident Evil: The Umbrella Chronicles (still trying to get the Darkside Chronicles)

Xbox 360: Gears of War 2 and Dead Rising (Assassin's Creed 2 is showing promise though, just not far enough to tell yet)

Gameboy/GBC: Wario Land: Super Mario Land 3 and Wario Land 3

GBA: Mario and Luigi: Superstar Saga, Sword of Mana, and Metroid Fusion ( couldn't pick from them)

DS: Phoenix Wright series and The World Ends With You

PSP: Crisis Core: Final Fantasy VII

iPod Touch: TNA Wrestling and Brothers in Arms (gave me the best value of the time, not going based on what I'm playing the most now.)

PC: Monkey Island series, with The Curse of Monkey Island being the best of them.

Arcade: The Simpsons Arcade Game, Time Crisis 2, House of the Dead series (still gotta play 4, so I can't pick a single favorite from there)

Flash: Sonny 2 and The Last Stand 2

I think that covers everything... still have a few systems I've never played before, so if I get around to playing them (especially the Dreamcast...) I'll put my favs here.
